Research/Clinical Interest
Title
Mechanisms of injury and repair during Acute Kidney Injury
Description
AKI is a common unavoidable consequence, occurring in nearly 2% of all hospitalized patients. Therapy for AKI has remained elusive over the past few decades, underscoring the urgent need for identification of potential therapeutic targets. Following injury, the cascade of events leading to endothelial dysfunction, tubular epithelial injury and activation of tissue-resident and infiltrating macrophages consists of coordinated action of cytokines/chemokines and reactive oxygen species. In this connection, we have previously demonstrated that expression of H-ferritin (FtH) in the kidney is imperative for protection against AKI. FtH is an anti-oxidant that catalyzes the conversion of the potentially toxic ferrous ion to ferric ion and allows for safe incorporation of iron into the ferritin shell and thereby attenuating participation of free iron in ROS generation. My current research efforts are aimed at determining the mechanisms of ferritin mediated cytoprotection and cross-talk between the tubules and resident/infiltrating immune cells during injury. Toward these efforts, we have generated cell-specific ferritin knockout mice to evaluate their effect on injury-mediated inflammation and recovery responses during sepsis and AKI. We elucidate differential regulation of the omics - transcriptome, proteome and metabolome to identify key pathways that are modulated during injury. These studies will provide significant and multi-faceted understanding of the major cellular components (tubular epithelium and macrophages) that are involved in the injury mediated responses following AKI and therefore, provide a potential repertoire of targeted therapeutics for AKI and other immunologically driven diseases.
